If they are raised (like little bumps) then you are looking at "tooth," which usually signals little pockets of flavor in the leaf.
If they are flat, with a white ring, or a green ring, or perhaps just a different shade (lighter or darker) of brown than the wrapper leaf, then it is probably just a "sunspot" on the leaf where a droplet of water caused a slight burn on the leaf in the sun. Either way, they are harmless (even sought after in the case of tooth). Cameroon & Sumatra are well known for having "tooth."
