Well, I think I've got it. After inspecting everything about 5 or 6 times over the last month, I finally noticed some cobwebs between the joists moving near the fan assembly.

I removed the ceiling tiles to discover that the flange on the assembly had bent back during installation. Probably half of what was being exhausted was going right into the basement.

45 minutes later, I was enjoying a smoke in my office - and there was no response from my wife! Smoked four more over the course of the weekend and she never mentioned a thing

I let the exhaust fan run for an hour after I'm done, then turn on the air purifier overnight. Nary an odor in the room. Thanks to everyone for your input, I am in smokers paradise!

Now on to renovating the room . . .
